yes you can put puppy and a virtual OS onto a pendrive... i think the only limitation is how big the pendrive is. For instance i can fit puppy and windows xp on my 4gb, although that doesn't leave much room..
to use virtual box you will need a CD or an iso image of the operating system that you want to install, click the new button in virtual box and follow the instructions , your OS will install as it would normally, but inside virtualbox.
to put it all on the pendrive you would use the universal installer in puppys menu to install puppy there..... then just run puppy and install your virtual OS
maybe try it on your harddrive first
give it a go and see
